cGAS

The cGAS enzyme recognizes foreign, intracellular double-stranded DNA and catalyzes the formation of the second messenger cyclic GMP-AMP which initiates a cascade ultimately activating transcription of Type I interferon genes. Human mutations in the cGAS pathway are associated with the neurodevelopmental disorder, Aicardi-Goutières syndrome, and overactivation of cGAS has been linked to Systemic Lupus Erythematosus. Ligand has identified novel potent inhibitors of cGAS which exhibit cellular activity and show similar potencies against human and murine forms of the enzyme.
